摘要:
通过建立测定磷霉素氨基葡萄糖盐原料药中有机溶剂乙醇和二氯甲烷残留量的顶空气相色谱的方法,对制备样品中的乙醇和二氯甲烷的残留量进行了检查。实验通过使用顶空气相色谱仪,对残留溶剂的系统适用性、线性范围、定量限、检测限、精密度和加标回收率进行考察,并且计算出了样品中乙醇和二氯甲烷的溶剂残留量。结果表明:乙醇,二氯甲烷的定量限分别为1.568 4和0.612 4 μg/mL,检测限分别为0.522 8和0.153 1 μg/mL;线性相关系数为0.999 8和0.997 9;2种有机溶剂的回收率均在99.2%~100.3%之间。本检测方法可适用于磷霉素氨基葡萄糖盐原料药中的有机残留溶剂的检查。
Abstract:
A headspace gas chromatography was established to determine the residues of organic solvents ethanol and dichloromethane in fosfomycin glucosamine active pharmaceutical ingredients, and the residues of ethanol and dichloromethane in the prepared samples were calculated. By using of the headspace gas chromatography, the system suitability, linearity range, limit of quantitation, detectability, precision and spiked recovery of residual solvent were investigated, and the solvent residues of ethanol and dichloromethane in the sample were calculated. The results showed that the limits of quantitation for ethanol and dichloromethane are 1.568 4?μg/mL and 0.612 4 μg/mL, the detectabilities are 0.522 8 and 0.153 1 μg/mL, and the linear correlation coefficients are 0.999 8 and 0.997 9, respectively. The recoveries of the two solvents are in the range of 99.2%-100.3%. The experimental results show that this method can be applied to the detection of organic residual solvents in fosfomycin glucosamine active pharmaceutical ingredients.
