摘要:
优化并建立液液萃取-气相色谱法测定糖精钠中的邻、对甲苯磺酰胺的残留量的检测方法。通过用水溶解糖精钠,加乙酸乙酯萃取其中的邻、对甲苯磺酰胺;并采用DB-17色谱柱进行气相色谱检测,100 ℃保持2 min,以10 ℃/min速率升至260 ℃,保持12 min。结果表明:邻甲苯磺酰胺在质量浓度4.04~40.37 μg/mL内,峰面积与浓度呈良好线性关系,平均回收率为104.9%,相对标准偏差为3.2%(n=12),检测限为0.807 μg/mL。对甲苯磺酰胺在4.06~40.61 μg/mL质量浓度内,峰面积与浓度呈良好线性关系,平均回收率为89.3%,相对标准偏差为1.5%(n=12),检测限为0.812 μg/mL。3个批次的糖精钠样品中邻、对甲苯磺酰胺含量均低于限度,符合药典相关要求。本方法优化了前处理工艺,操作简便,灵敏度与准确性高,无环境污染,可用于糖精钠中甲苯磺酰胺残留量检测。
Abstract:
To determine o-toluenesulfonamide and p-toluenesulfonamide in saccharin sodium, a liquid-liquid extraction coupled with gas chromatography method was established and optimized. The saccharin sodium sample was dissolved in water, and the o- and p-toluenesulfonamide impurities were extracted with ethyl acetate. A DB-17 chromatographic column was adopted for gas chromatographic detection. The column temperature was initially maintained at 100 ℃ for 2 min, then raised to 260 ℃ at a heating rate of 10 ℃/min and held constant for 12 min. Results showed that o-toluenesulfonamide exhibited a good linear relationship between peak area and concentration within the range of 4.04-40.37 μg/mL, with an average recovery of 104.9%, a relative standard deviation (RSD) of 3.2% (n = 12), and a limit of detection of 0.807 μg/mL. A favorable linearity was achieved for p-toluenesulfonamide in the range of 4.06-40.61 μg/mL, with an average recovery of 89.3%, an RSD of 1.5%(n = 12), and a limit of detection of 0.812 μg/mL. The contents of o- and p-toluenesulfonamide in three batches of saccharin sodium samples were all below the detection limits, which complied with the pharmacopoeia requirements. With optimized sample pretreatment procedures, the proposed method features simple operation, high sensitivity and accuracy, as well as environmental friendliness. It is suitable for routine detection of toluenesulfonamide residues in saccharin sodium.
