摘要:
比较研究了两个茯苓菌株的菌丝生长速度,比较了固体接种、液体接种对茯苓菌丝体生物量的影响,采用正交试验探究了不同碳源浓度、不同氮源浓度和不同初始pH值对茯苓菌丝生物量和茯苓发酵液中胞外多糖的影响.结果表明:其作用大小是碳源>氮源>初始pH值.以菌丝体干重为指标,茯苓液体发酵的最适发酵培养基为:碳源(葡萄糖)3%,氮源(酵母浸膏0.51%,蛋白胨0.4%),K2HPO4 0.1%, KH2PO4  0.046%,MgSO4·7H2O 0.05%,水1L,最适初始pH值5.5;以多糖为指标,茯苓液体发酵的最适发酵培养基为:碳源(葡萄糖)3.5%,氮源(酵母浸膏0.637%,蛋白胨0.5%),K2HPO4 0.1%, KH2PO4  0.046%,MgSO4·7H2O 0.05%(以上百分数均为质量分数),水1 L,最适初始pH值5.0.
Abstract:
The mycelia growth rate of two Poria cocos strains were measured. The effect of solid spawn and liquid spawn on biomass of P. cocos was studied. The optimum liquid fermentation mediums were studied with the mycelia dry weight and the contents of exopolysaccharide as markers by orthogonal test. The result showed that the ranking of the influence on fermentation was as follows : carbon sources > nitrogen sources > initial pH. The optimum mediums were as follow:3.0% or 3.5% glucose,0.51% or 0.637% yeast extract,0.4% or 0.5% peptone,0.1% K2HPO4,0.046% KH2PO4, MgSO4·7H2O 0.05%, the optimum initial pH 5.5 or pH 5.0.
