对一株命名为st10新产ε-聚赖氨酸菌株(ε-Polylysine )进行测序,产物鉴定,及用响应面法确定了该菌株的最优摇瓶发酵配方。对st10菌株的16s rRNA比对确定该菌株属于白色链霉菌属,通过特异颜色反应、透明圈现象、薄层层析及电泳方法证明了产物为ε-聚赖氨酸,其分子量在5 000 Da以下。单因素实验发现,适合菌株发酵的碳源和氮源分别为甘油,酵母粉和硫酸铵,其中硫酸铵利于降低发酵液pH和酵母粉促进菌量大量繁殖,均有利于ε-聚赖氨酸的分泌。响应面实验结果表明:最适ε-聚赖氨酸分泌的配方为酵母粉4.05 g/L,硫酸铵7.63 g/L,甘油53.26 g/L,实际产量为0.724 g/L。该成果对ε-聚赖氨酸表达制备,发酵罐发酵及菌株改造等后续研究奠定了基础。
Abstract:
In this paper, a new ε-polylysine-producing strain (named st10) was sequenced, identified, and the optimal fermentation formula was determined by response surface methodology. The strain st10 was identified as streptomyces albus by 16s rRNA comparison. Result of specific color reaction, transparent ring phenomenon, thin layer chromatography and electrophoresis showed that the product is polylysine, and its molecular weight is below 5 000 Da. Single factor experiments showed that glycerol, yeast powder and ammonium sulfate are the most suitable carbon and nitrogen sources for fermentation, respectively. The presence of ammonium sulfate is conducive to decrease the pH value of fermentation broth and the yeast powder promotes the growth of bacteria, both of which are favorable for the secretion of polylysine. Response surface analysis indicated that the optimal formulation for polylysine secretion is yeast powder 4.05 g/L, ammonium sulfate 7.63 g/L, glycerol 53.26 g/L, and the actual yield is 0.724 g/L. The work laid the foundation for polylysine production measurements, fed-batch fermentation studies in the fermenters and other following up studies.
